当分野は日本で最初のDepartment of Neuroimmunologyである。 主宰の錫村は、現在国際的なスタンダードとなっているミクログリア単離培養法(’shaking off’ method)を世界に先駆けて確立したパイオニアの一人である。 神経系は永く免疫学的特権部位とよばれ、免疫系の監視から免れる特異な部位と考えられてきた。しかしながら、神経系もある種の条件下では免疫学的特権部位でなくなり、自己免疫機序による疾患群が存在する。これらの特殊な状況下での神経系における免疫反応、病態発現機序、治療法の開発を研究する学問が神経免疫学である。 これまでに我々は、中枢神経系ではミクログリアやアストロサイトなどのグリア細胞が活発にサイトカイン/ケモカインを産生し、免疫系とは異なった独自のサイトカイン/ケモカインネットワークを形成していること、グリア細胞(主としてミクログリア)が抗原提示細胞としてT細胞に抗原提示をし、同時にヘルパーT細胞の分化を規定することにより神経系での免疫病態を調節している事を明らかにしてきた。 さらに近年、アルツハイマー病、筋萎縮側索硬化症、ハンチントン病、多発性硬化症、虚血性脳疾患などの様々な神経変性疾患において、ミクログリアが病態に重要な役割を果たしていることが明らかとなっている。 我々は、活性化したミクログリアが、グルタミン酸、一酸化窒素、炎症性サイトカインといった各種傷害因子の放出を介して、神経変性にも大きく関与している事を明らかにしてきた。 さらに、種々の外的ストレスが自律神経や内分泌系を介して免疫系の調節していることも明らかになっており、精神的ストレスが内分泌系や交感神経を介して末梢の免疫細胞の機能変化を誘導し,自己免疫の発症の誘因になることも明らかになっている。これら神経-免疫-内分泌系の研究も神経免疫学の重要な課題である。 当教室の研究目標 今年度のプロジェクト研究(他施設との共同研究を含む) 1. 神経変性に対する革新的治療薬の研究開発
